Equilibrium dialysis-:
Equilibrium dialysis : The method of equilibrium dialysis can be used to study the interaction between drug and protein or between metal ions and other macromolecules . A number of sacs of dialyzing membrane containing the same concentration of the protein ( or other macromolecule ) are suspended in the drug ( or metal ion ) solutions of various concentrations Fig . 3.6 . The medium on both the sides of the membrane is same . The equilibrium is allowed to attain by leaving the assembly undisturbed for nearly 24 hours . The concentration of free drug on both the sides is determined by analysing the inner and outer solutions . The dialysing membrane allows the transfer of the small drug molecules ( or the metal ions ) , however , hinders the passage of the protein ( or macromolecule ) . If there is no complexation , the concentration of the drug on both the sides is the same . If the drug after diffusing through the membrane complexes with the protein in the sac , the drug concentration ( total ) in the sac will be greater than the concentration outside .
